DNA/RNA Size QC

This service will determine the size profile of RNA or DNA samples, including primary biological samples and NGS libraries or pools.

The size profile of different sample types indicates:

  • RNA integrity (degree of degradation), reflected in the RQN value. RQN>7 is considered intact.
  • The average size for NGS libraries, which can be used to calculate the molarity.
  • The average size of genomic DNA samples.

Based on the concentration and expected size distribution of your samples, select the appropriate Agilent instrument for your project when placing an order.

Fragment Analyzer – used for most samples

 Size RangeConcentration
DNA Samples50bp – 1.5 Kb0.25 - 5ng/ul
RNA Samples200nt – 6 Kb1 - 5ng/ul
(recommended: 3 - 5ng/ul for best detection

Femto Pulse – used to determine the size of high molecular weight genomic DNA (1.3kb to >100kb) and for very low concentration RNA samples (< 1ng/ul).

The results will include the size distribution profile as well as quantitative metrics for called peaks. The reported sample concentration may not be accurate and a Qubit or Nanodrop/Quickdrop reading is the preferred method to determine sample concentration. Samples containing salts or other impurities that affect electrophoretic mobility may not have accurate sizing with this assay.
